Different Cell and Tissue Behavior of Micro-/Nano-Tubes and Micro-/Nano-Nets Topographies on Selective Laser Melting Titanium to Enhance Osseointegration

Background and purpose: Micro-/nano-tubes (TNTs) and micro-/nano-nets (TNNs) are the common and sensible choice in the first step of combined modifications of titanium surface for further functionalization in the purpose of extended indications and therapeutic effect. It is important to recognize the respective biologic reactions of these two substrates for guiding a biologically based first-step selection.

Materials and methods: TNTs were produced by anodic oxidation and TNNs were formed by alkali-heat treatment. The original selective laser melting (SLM) titanium surface was set as control. Surface characterization was evaluated by scanning electron microscopy, surface roughness, and water contact angle measurements.

Osteoclastogenesis and osteogenesis were measured. MC3T3-E1 cells and RAW 264.7 cells were used for in vitro assay in terms of adhesion, proliferation, and differentiation. In vivo assessments were taken on Beagle dogs with micro-CT and histological analysis.

Results: TNN and TNT groups performed decreased roughness and increased hydrophilicity compared with SLM group. For biological detections, the highest ALP activity and osteogenesis-related genes expression were observed in TNT group followed by TNN group (P <0.05).

Interestingly, when it comes to the osteoclastogenesis, TNNs displayed lowest TRAP activity and osteoclastogenesis-related genes expression and TNTs were lower than SLM but higher than TNNs (P <0.05). BV/TV around implants was highest in TNT group after 4 weeks (P <0.05). HE, ALP and TRAP staining showed that osteogenic and osteoclastic activity around TNTs were both higher than TNNs (P <0.05).

Conclusion: TNNs and TNTs have dual advantages in promotion of osteogenesis and inhibition of osteoclastogenesis. Furthermore, TNNs showed better capability in inhibiting osteoclast activity while TNTs facilitated stronger osteogenesis. Our results implied that TNT substrates would take advantage in early application after implantation, while diseases with inappropriate osteoclast activity would prefer TNN substrates, which will guide a biologically based first-step selection on combined modification for different clinical purposes.

How does a non-optimal tube potential influence radiation dose to the patient in lumbar spine radiography?

Introduction: When comparing the radiation dose to the patient, the lumbar spine has one of the highest dose values in general radiography, therefore the procedure needs to be optimised. The aim of this study was to investigate the effect of a non-optimal tube potential (66 kV) during anteroposterior (AP) lumbar spine radiography on the radiation dose received by the patient compared with the radiation dose when an optimal tube potential (79 kV) is used, in accordance with European guidelines.

Methods: This retrospective study involved 100 patients referred for lumbar radiography in two different diagnostic departments. Half of the patients were admitted to a department which used optimal tube potential and the other half to the department which used non-optimal tube potential for AP lumbar spine radiography protocols. The height and weight of the patients were collected to calculate the body mass index (BMI) of the patients. The image field size and dose area product (DAP) values were collected after each imaging session. The effective dose and selected organ dose were calculated using the PCXMC 2.0 program.

Results: The results showed that a non-optimal tube potential resulted in a significant increase in the DAP value by 360% (p < 0.001) and a significant increase in the effective dose by 160% (p < 0.001). Dose to selected organs due to non-optimal tube potential increased from 107% (breasts) up to 631% (prostate) (p < 0.001). The images were not assessed using visual grading characteristics (VGC) analysis, but the radiologists evaluated all the images appropriate for diagnostic reading.

Conclusion: Based on our study’s stated results, we can conclude that optimal tube potential use is essential to achieve the ALARA principle.

Implications for practice: The study shows the effect of a non-optimal tube potential on the radiation dose received by the patient during radiography of the lumbar spine. This could influence possible diagnostic departments to consider protocol optimisation due to the high radiation dose received by the patient.

Adenomatoid Tumor of Fallopian Tube With “Deciduoid” Morphology and Atypical Nuclear Features

Adenomatoid tumor is a benign neoplasm of mesothelial origin. Adenomatoid tumor in female genital tract shows typical morphologic features with bland nuclei. Deciduoid morphology has not been reported in adenomatoid tumor. Tumors with deciduoid cells and atypical nuclear features may pose a diagnostic challenge and raise the suspicion of malignancy.

We present a case of fallopian tube adenomatoid tumor with deciduoid morphology and atypical nuclear features in a 39-year-old woman with prolonged progestin therapy. We hypothesize that the unusual morphological changes in adenomatoid tumor, like deciduoid morphology and nuclear atypia, may be secondary to hormone effects.

 

PARP1 Is Targeted by miR-519a-3p and Promotes the Migration, Invasion, and Tube Formation of Ovarian Cancer Cells

Background: Poly ADP-ribose polymerase 1 (PARP1) has been discovered to be implicated in ovarian cancer (OC), but its interaction with microRNA (miR)-519a-3p remained poorly understood. This study aimed to uncover their roles and interactions in OC.

Materials and Methods: Clinical tissue from OC patients and adjacent normal tissue were collected, and the survival rates of OC patients with high or low PARP1 expression were analyzed by Kaplan-Meier curve. After transfection, OC cell viability, migration, and tube formation were detected with cell counting kit-8 (CCK-8) assay, scratch assay, and tube formation assay, respectively.

Target gene of miR-519a-3p and potential binding sites between them were predicted with TargetScan and confirmed using dual-luciferase reporter assay. Relative expressions of miR-519a-3p, PARP1, E-cadherin, N-cadherin, SNAIL, vascular endothelial growth factor (VEGF), and p53 were measured by quantitative real-time polymerase chain reaction and Western blot as needed.

Results: PARP1 expression was upregulated in OC, which was related to poor prognosis of OC patients. Silencing PARP1 decreased PARP1 expression and suppressed viability, migration, invasion, and tube formation in OC cells, while overexpressed PARP1 did the opposite. PARP1 was the target gene of miR-519a-3p, and it reversed the effects of miR-519a-3p on the migration, invasion, and tube formation of OC cells by upregulating the expressions of PAR, PARP1, N-cadherin, SNAIL, and VEGF and downregulating those of E-cadherin and p53.

Conclusion: PARP1, a target gene of miR-519a-3p, promoted the migration, invasion, and tube formation of OC cells, providing a possible therapeutic target for treatment of OC patients.
